**[212Pb]VMT01 + nivolumab** is an investigational combination therapy for melanoma comprised of two agents: [212Pb]VMT01 and nivolumab. - **[212Pb]VMT01** is a targeted radiopharmaceutical that consists of a proprietary low-molecular-weight peptide (VMT01) labeled with lead-212, an alpha-emitting isotope. VMT01 binds to the melanocortin 1 receptor (MC1R), which is overexpressed on melanoma cells, delivering targeted alpha-particle radiation to tumor cells to induce tumor cell death. - **Nivolumab** is a monoclonal antibody immune checkpoint inhibitor targeting programmed cell death protein 1 (PD-1); it enhances anti-tumor immunity by preventing PD-1 mediated inhibition of T-cell activity. This combination is under clinical investigation for patients with unresectable or metastatic melanoma who are MC1R-positive by imaging. The rationale for the combination is the potential synergistic effect of localized alpha-particle-induced tumor cell death (immunogenic cell death) with immune activation via checkpoint inhibition (PD-1 blockade). The two-drug regimen is in phase 1/2a development with both monotherapy and combination arms. Perspective Therapeutics sponsors the trial, nivolumab is supplied by Bristol Myers Squibb[2][4][5][1].
